When living cells are studied wet mount has following advantages over dry mount:

  • Image quality is improved due to refractive index of water
  • It is possible to observe real time cell functions like feeding and division.
  • Organisms are viewed in their natural form and colour as dry mount can fade or change the colour of the organism

Oak leaf can't be used in wet mount investigation because it is too thick. It blocks light coming from the stage and observer cant observe the specimen's details.
